PUREBRIGHT MA-P12-vc-pAB-NP is a cleavable pendant-type heterobifunctional monodisperse PEG linker designed for antibody-drug conjugates (ADCs), featuring two monodisperse PEG chains, two distinct functional groups a maleimide and a p-nitrophenyl carbonate—and a lysosomal enzyme-cleavable dipeptide unit combined with a self-immolative moiety, enabling controlled intracellular drug release under lysosomal conditions.
The pendant-like structure forms a hydrophilic domain that masks the hydrophobicity of payloads, enhancing aqueous solubility and improving pharmacokinetics. In vivo and in vitro studies demonstrated that ADCs prepared with MA-P12-vc-pAB-NP showed reduced aggregation, improved tolerability, extended circulation half-life, and enhanced anti-tumor efficacy compared to conventional non-PEGylated linkers.
