Abstract
Nucleic acid vaccines, DNA and messenger RNA (mRNA), have emerged as promising modalities for infectious disease and for cancer immunotherapy due, in part, to shortened producting cycles and high potency. Lipid nanoparticles (LNP) composed of ionizable lipids are important components of such vaccines as they can convey and present the nucleic acid effectively to the immune system. However, toxicity of ionizable lipids and the challenges of cold chain logistics of LNP have limited their clinical use. Further development of LNP requires 1)reduced systemic toxicity, 2) improved endosomal escape, 3) potent and cytotoxic T-cell specific adjuvating function, 4) reduced adverse reactions, 5) improved storage stability of LNP.
