For site-specific delivery, lipid nanoparticles (LNPs) modified with targeting ligands are widely used in advanced drug delivery systems. SUNBRIGHT GS-020AZ is a functionalized PEG lipid designed for efficient ligand conjugation in LNP formulations. It features two C18:0 fatty acids (stearic acid) connected at the 1- and 2-positions of a glycerol backbone, with a 2,000 Da polyethylene glycol (PEG) chain at the 3-position, terminated with an azide group. The azide group enables site-specific conjugation with strained alkyne derivatives such as cyclooctyne (DBCO or BCN)-modified molecules (such as peptides, antibodies, or other ligands) through copper-free click chemistry, forming a stable triazole linkage. This facilitates precise cellular targeting and enhances formulation versatility. GS-020AZ does not have a charge that could adversely affect the stability of LNPs and is useful for developing active targeting delivery systems.
